Same problem but was able to get it to work by un-ticking the "Ignore NTFS junction points (reparse points)" option as described.
1. Will this be the new default for profiles or will we have to remember to do this?
2. What other (expected) effect does un-ticking it have?

In my case, given some performance issues with the option un-ticked (discussed in this thread), I've decided to turn off the "new" files on demand option of OneDrive, and SBF returned to what it was. It's fine again.

This does not work. It will then give you an error stating "Failed to prepare <left side name>: The drive <unintelligible character> does not exist!"

You need to un-tick the option "Ignore NTFS junction points (reparse points)" when backing-up files to/from local OneDrive folder.1. Will this be the new default for profiles or will we have to remember to do this?
If you un-tick that option, SyncBack scans the target directory a Junction point is pointing to. By default, SyncBack will ignore junction points.2. What other (expected) effect does un-ticking it have?

Wouldn't it be wise to have this setting (unchecked) as a default?
Most people probably don't discriminate between normal folders and NTFS reparse points and expect SB to backup the latter with any basic profile setup from default.
I myself am a good example, I stumbled upon this issue when searching for expected backups from a source that contained also my OneDrive folder. I found OneDrive excluded from the backup. Luckily I could recover it from a sharing partner. I am not a newbie to SB (using SB SE) but still didn't notice this quite important setting.
